Hydrogen peroxide has been used in the bleaching of cotton and other textiles. It has been also used for cosmetic purposes and as a rocket propellant. This colorless, corrosive substance reacts with certain organic compounds to form crystalline solids. The reaction is very reactive and it can cause damage to the fabric. Typically, hydrogen peroxide is produced as an aqueous solution of different strengths. Usually, solutions containing more than 8 percent hydrogen peroxide are toxic and corrosive to the skin.

Sodium perborate, commonly used as a mild oxidizing agent, is used in chlorine-free bleach products. Ascorbate plays an important role in the production of hydrogen peroxide. Moreover, many chelating agents are also available which can bind heavy metal ions. Using these ingredients together can improve the functionality of unbleached cotton, which has been considered a potential wound dressing material.

In this study, a variety of copper/ascorbate formulations were designed to modulate the level of hydrogen peroxide and its antibacterial activity for wound healing. In addition, the relative roles of these ingredients were elucidated to understand the interaction of these two agents with greige cotton.

A comparison of the hydroxyl radical generation in both the adsorbed and in situ synthesized nanoparticles was made. It was observed that the in situ formation of Cu+/Asc resulted in a 20 micromolar hydrogen peroxide generation, which was five-fold lower than the adsorbed formulation. Similarly, the adsorbed Cu+/Asc formulation had a lower hydrogen peroxide activity wash-out.

Increasing the concentration of the copper/ascorbate formulation on the cotton fiber exhibited optimal hydrogen peroxide generation. Furthermore, higher concentrations of the copper/ascorbate on the fiber were found to promote the formation of hydrogen peroxide, which is consistent with the role of these materials in the oxidation of ascorbic acid.

However, the antibacterial activity of the copper/ascorbate-adsorbed cotton was limited. The presence of hydroxyl radicals in the fiber could be due to polyphenolic autoxidation, which occurs in the fiber, or superoxide dismutase dismutation, which may happen in pectin hydrolysis. Therefore, the titer of these compounds was determined using ICP analysis.

Thus, the results showed that both copper and ascorbate could produce hydrogen peroxide when applied to greige cotton, with an increased effectiveness of hydrogen peroxide generation when increasing the amount of copper. Consequently, greige cotton-based formulations may be used to enhance the peroxide generation in wounds.
